摘要
目的:探讨γ线复合微波辐射对HL-60细胞增殖分化能力的影响及意义。方法:HL-60细胞随机分为对照组、微波辐射组、γ线辐射组、γ线复合微波辐射组,细胞经5.5 Gy60Coγ线辐射后立即进行微波辐射(平均功率密度50 mW/cm^2、辐射时间5 min),于辐射后即刻、6 h1、2 h和24 h,应用MTT、3H-TdR掺入、硝基蓝四氮唑(NBT)还原结合形态学观测HL-60细胞增殖活性、DNA合成及细胞分化能力的改变。结果:与对照组比较,微波辐射组于辐射后12-24 h HL-60细胞增殖活性和DNA合成明显降低,NBT还原反应阳性的细胞数明显减少;γ线辐射组和复合组于辐射后6-24 h细胞增殖活性和DNA合成显著降低,NBT还原反应阳性的细胞数显著减少,且复合组变化较γ线辐射组更明显。结论:γ线和复合微波辐射明显抑制HL-60细胞增殖分化,其损伤以γ射线为主,微波可加重其效应。
Objective:To investigate the effect of γ-ray combined with microwave radiation on proliferation and differentiation potential of HL-60 cells and its significance.Methods:HL-60 cells were divided into normal control,microwave radiation,γ-ray radiation,γ-ray combined microwave radiation groups espectively,which was irradiated by microwave with average power density 50 mW/cm2 for five minutes before exposure to 5.5 Gy 60CO γ ray.Changes of cell proliferation,DNA synthesis and cell differentiation potential of HL-60 cells were detected by MTT,3H-TdR incorporation,NBT reduction and morphology observation at 0 h,6 h,12 h and 24 h after radiation.Results:Compared with normal testis,there was a obvious decrease in cell proliferation and DNA synthesis of HL-60 cells at 12 h and 24 h after radiation,the cell population with positive NBT reduction reaction was also obviously decreased in microwave radiation group.The cell proliferation and DNA synthesis of HL-60 cells was decreased significantly in the γ-ray radiation and γ-ray combined microwave radiation groups from 6 h to 24 h,the cell population with positive NBT reduction reaction was also significantly decreased,which of the γ-ray combined microwave radiation group was more obvious than that of the γ-ray radiation group.Conclusion:The γ-ray combined microwave radiation inhibits obviously proliferation and differentiation of HL-60 cells,which is mainly caused by γ-ray,microwave radiation may exert aggravation on its injury effect.
