摘要
利用马尾松未成熟胚为外植体,在预备实验的基础上,通过对不同时期不同家系马尾松幼胚的采集、接种,诱导产生胚性愈伤组织,计算实验数据,利用SPSS13进行数据分析。结果表明:以LP为基本培养基,最易诱导出胚性愈伤组织的采种时期是4月中旬至5月下旬;在这一时期不同家系对诱导率影响的差异不显著;随着幼胚成长,胚性愈伤组织的诱导率也逐步下降;马尾松幼胚的胚性愈伤组织诱导过程需在暗培养条件下进行;在初步试验中,诱导胚性愈伤组织最佳的是第③号激素处理。
Based on the preliminary experiments,we collected the immature embryos of Pinus massoniana from 3 seed trees at different periods,then inoculated them in batches according to the differences of seed trees and collected-periods on the LP media;calculated and analysed the inductivities of embryonic callus with SPSS13 software.The results show that The most suitable time to induce the embryonic callus was from mid-April to late May,in this period the differences between families was nonsignificant.The inductivities were declining step by step with the maturing of the immature embryos;the embryonic callus induction of the immature embryo of Pinus massoniana should be cultivated in the dark.In the primary test,the best hormone treatment was the number three media.
