摘要
吲哚胺2,3-双加氧酶(IDO)是一种含铁血红素单体蛋白,其表达和活化可以被干扰素γ强烈诱导。通过催化色氨酸降解,使表达IDO的肿瘤细胞周围形成低色氨酸环境,由于色氨酸是体内必需氨基酸,缺乏色氨酸时蛋白质合成下降,外周T淋巴细胞的增殖受到抑制。目前大量研究表明IDO在白血病细胞中较高表达,抑制T细胞的增生和T细胞介导的免疫反应,使T细胞活化信号转导受阻,诱导白血病细胞发生免疫逃逸。
Indoleamine 2,3-dioxygenase(IDO)is a monomer heme iron-containing protein,whose expression and activation can be potently induced by IFN-γ.IDO catalytic degradation,tryptophan,forms a low tryptophan environment surrounding tumor cells,which express IDO.The reduced protein synthesis,due to the lack of tryptophan,an essential amino acid,will suppress the proliferation of peripheral T lymphocytes.Extensive studies have shown that the highly expressed IDO in leukemic cells inhibite the proliferation of T cells and T cell-mediated immune reaction,which handicaps the signal transduction of T cell activation and induces the immune escape of leukemic cells.
