消毒方法和培养基对青钱柳茎段腋芽萌发的影响 被引量:14

摘要 为建立有效的青钱柳组培快繁体系,以青钱柳茎段腋芽作为外植体,研究了不同消毒方法和基本培养基对腋芽萌发的影响。结果表明,先用70%酒精消毒30 s,再用0.1%氯化汞消毒12 min,腋芽污染率最低、芽萌发率最高,分别为31.25%、81.82%。极差分析结果表明,酒精处理时间对萌发率影响较大,是诱导腋芽萌发的主要因素,而氯化汞处理时间对污染率影响较大,是影响消毒效果的主要因素。在所试的7个基本培养基中,芽萌发率存在一定的差异,以WPM和改良MS培养基效果最佳,萌发率分别达到83.3%和75.76%,并且与其他处理差异显著(P<0.05);WPM培养基芽生长状况最好,叶深绿色、叶片伸展、茎粗壮。研究结果为青钱柳组培快繁体系的建立提供依据,并为其他林木的快繁研究提供技术参考。 In order to set up the high efficient tissue culture and rapid propagation system of Cyclocarya paliurus, axillary buds from stem segments were used as explant in this paper. Effects of the different disinfection methods and the basic culture medium component on its germination of axillary buds were studied . The results showed that the time of 70% alcohol disinfection is 30 seconds and the time of 0.1% HgCl2 disinfection is 12 minutes, which the lowest polluting rate was 31.25% and the highest germinating rate of the bud was 81.82%. Range analysis results showed that the effect of time of alcohol treatment on germinating rate was bigger,which was the main factor of inducing germination of axillary buds. But the effect of HgCl2 treatment time on pollution rate was bigger, which was the main factor of disinfection effect. In the 7 basic culture medium, germinating rate of the bud show some difference. WPM and modified MS were the optimal culture medium ,which their germination rate were 83.33% and 75.76% respectively and the difference was significant compared with the other treatments( P 〈 0.05 ). Growth condition of buds was the best under WPM culture medium condition, whose leaves were deep green and expansion, and stem were stronger. The results provided basis for seting up the tissue culture and rapid propagation of Cyclocarya paliurus, and provided technical reference for the study of rapid propagation of other forestry trees.
