摘要
为简化定向进化筛选环己酮还原能力提高的突变型糖多孢红霉菌聚酮合成酶酮还原酶域的繁琐工作,建立了利用环己酮和2,4-二硝基苯肼的显色反应来检测环己酮含量的方法。环己酮浓度在0~5mg·mL-1范围内与吸光度线性关系良好,线性相关系数为0.9973。在10mL转化液的实验条件下,方法的检测限为188μg.L-1,相对标准差为1.52%。该方法操作简便、灵敏度较高、结果可靠,能满足筛选突变体的要求。
In order to simplify the screening of the mutant ketoreductase domain in the first module of polyketide synthase from Saccharopolyspora erythraea which had increased ability of cyclohexanone reduction,a determination method of cyclohexanone content was established by making use of the reaction between cyclohexanone and 2,4-dinitrophenylhydrazine.The method offered good working curve linearity within 05 mg·L-1,correlation coefficient was 0.9973,the relative standard deviation was 1.52% and the detection limit was 188 μg·L-1 under the condition of 10 mL ferment.This result showed that the method was simple,accurate,sensitive and fit for screening the mutants.
