人牙本质磷蛋白的提取与特性分析 被引量:4

摘要 目的建立人牙本质磷蛋白(dentinphosphoprotein,DPP)的提取方法并分析其特性。方法将人牙本质粉用0.6mol/LHCl脱矿后,用DEAESepharoseCL6B离子交换层析柱分离纯化DPP,取纯化的人DPP,用紫外分光光度计检测其吸光度值;用等离子发射光谱分析其磷含量;SDSPAGE测量相对分子质量;并在水解后进行氨基酸含量分析。结果用0.6mol/LHCl脱矿,可以有效地提取人DPP。所提取的DPP是磷含量较高的蛋白质。相对分子质量分别为:141000,124000,108000,氨基酸组成中天门冬氨酸和丝氨酸的含量较高,分别为23.8%和19.1%。结论用0.6mol/LHCl脱矿和离子交换层析等方法可以分离纯化人DPP;人DPP是牙本质中一组含磷量较高、相对分子质量较大(100000~150000)、富含丝氨酸和天门冬氨酸的阴离子蛋白质。
