抗增殖基因BTG2在人胰腺癌组织中的表达及其抗增殖作用

摘要 目的 研究BTG2(B cell translocation gene2)在人胰腺癌中的表达以及对胰腺癌细胞生长的作用。方法利用RT-PCR法研究31例人胰腺癌及同例癌旁组织BTG2mRNA的表达。利用免疫沉淀法检测其BTG2蛋白的表达。用pcDNA3.1-BTG2质粒转染人胰腺癌细胞株sw1990,用MTT法测定质粒转染后sw1990/pcDNA3.1.BTG2细胞生长曲线;应用AnnexinV-FITC凋亡检测试剂,用流式细胞仪测定sw1990/pcDNA3.1.BTG2细胞凋亡的情况。结果人胰腺癌BTG2mRNA的表达明显低于癌旁正常组织的表达(P〈0.05);人胰腺癌BTG2蛋白的表达低于癌旁正常组织的表达(P〈0.01);pcDNA3.1-BTG2质粒的转染能明显抑制胰腺癌细胞株sw1990的生长。结论BTG2基因在人胰腺癌中的表达下调可能与其恶性行为有关;BTG2基因的过度表达可通过诱导凋亡而抑制人胰腺癌细胞株sw1990的生长。BTG2可能是胰腺癌基因治疗的一个靶点。 Objective To Explore the expression of BTG2 (B cell translocation gene 2) in human pancreatic cancer. And its correlation with the proliferation of pancreatic cancer cells. Methods 31 pairs of pancreatic cancer and the tumor surrounding tissue were selected to detect the BTG2 mRNA and protein by RT-PCR and immunoprecipitation method respectively. Plasmid pcDNA3.1-BTG2 was introduced into human pancreatic cancer cell lines sw1990 by transfection. The effect of pcDNA3.1-BTG2 on sw1990 cell growth curve was estimated by M'VF method; Flow cytometry were used to measure the apoptosis index of rtansfected sw1990cells with Annexin V-FITC kit. Results The expression of BTG2 mRNA was distinctly suppressed in tissues than the tumor surrounding tissue (P〈0.05); as well as the BTG2 protein did (P〈0.05). The transfection of plasmid pcDNA3.1-BTG2 could significantly inhibit the growth of sw 1990 cells, and the apoptosis index of sw1990/pcDNA3.1-BTG2 has statistically meaning, significantly elevated upto 90% than that of control. Conclusions The suppression of BTG2 expression was probably related with the character of; the overexpress of BTG2 could inhibit the growth and inducing obviously apoptosis of sw1990. That implies that BTG2 maybe a new target for gene therapy of pancreatic cancer.
