背景:前期研究发现半乳凝素(galectin)-1高表达于结直肠侧向发育型肿瘤(KST)细胞株LST-R1,由此推测其可能与LST病变的侧向生长有关.目的:探讨galectin-1表达下调对LST-R1细胞侵袭表型的影响.方法:以RNA干扰(RNAi)技术沉默LST-R1细胞的galectin-1基因,采用异质性黏附实验、同质性黏附实验、趋化运动实验、软琼脂集落形成实验比较转染galectin-1短发夹RNA(shRNA)表达载体的p-shRNA1-LST细胞、转染空载体的p-LST细胞和对照组LST-R1细胞侵袭表型的差异.结果:p-shRNA1-LST细胞的异质性黏附细胞数增加,同质性黏附细胞数减少,趋化运动能力增强,细胞集落形成数增多.结论:galectin-1表达下调可致LST-R1细胞出现侵袭相关表型,提示galectin-1可能参与了LST病变浅表性扩散特性的形成.
Background: Galeetin-1 is overexpressed in colorectal laterally spreading tumor (LST) cell line LST-R1 in prophase research, and was presumed to be associated with the laterally spreading of LST lesion. Aims: To assess the influence of galectin-1 downregulation on the invasive phenotype of LST-R1 cells. Methods: RNA interference (RNAi) was employed to knock-down the expression of galectin-1 gene in LST-R1 cells. Invasive phenotypes, including heterotypic and homotypic cell adhesion, chemotactic motion and colony formation ability in soft agar of p-shRNA1-LST cells (transfected with galectin-1 short hairpin RNA expression vector), p-LST cells (transfected with blank vector), and controlled LST-R1 cells were assessed. Results: p-shRNA1-LST cells had more heterotypic adhesion cells, less homotypic adhesion cells, increased chemotactic motion and colony formation ability in soft agar. Conclusions: Downregulation of galectin-1 can induce the invasive phenotype of LST-R1 cells, which indicates that galectin-1 may play a role in developing the superficial spreading specificity of LST lesion.
