摘要
目的研究不对称二甲基精氨酸(ADMA)对THP-1源巨噬细胞以及其形成的泡沫细胞内胆固醇含量的影响.探讨其促使动脉粥样硬化发生的可能机制.方法以THP-1来源的巨噬/泡沫细胞模型为研究对象,用不同浓度ADMA在不同时间内进行体外干预;采用酶偶联比色法测定干预前后细胞内胆固醇含量的变化.结果 ADMA在体外以浓度和时间依赖的方式上调巨噬/泡沫细胞内总胆固醇的含量,增强巨噬/细胞吞噬脂质的能力.结论 ADMA增加巨噬/泡沫细胞内胆固醇含量,使过多的胆固醇在细胞内聚积,从而加重细胞泡沫化的程度.
Objictive To explore the effect of asymmetric dimethylarginine(ADMA) on cholesterol content in cultured THP-1 monocyte-derived macrophages and foam cells,so as to reveal the possible mechanism of ADMA in arteriosclerosis.Methods THP-1 derived macrophages and foam cells were incubated with ADMA at different concentrations for different times in vitro.Then enzyme-linked colorimetry assay was used to determine the influence of ADMA on intracellular cholesterol content before and after treatments.Result ADMA increased intracellular cholesterol content in THP-1 derived macrophages in a concentration and time dependent way in vitro,suggesting that ADMA promoted the phagocytosis of lipids in macrophages and foam cells.Conclusion ADMA can increase intracellular cholesterol content,and thus fasts the speed of macrophage-derived foam cells formation,and play a role in the development and progression of atherosclerosis.
