摘要
目的:筛选IL-2靶向脂质体的最佳处方,并对5-氟尿苷(5-fluorouridine,5-FUR)前药进行含量测定。方法:应用正交实验设计选择最佳处方配比;采用紫外分光光度法测定5-FUR前药的含量,检测波长为267nm。结果:筛选最佳处方:药脂比为1:10、磷脂与胆固醇的比例为3:1。5-FUP前药对照品浓度在0.002-0.02mg/ml的范围内与吸收度线性关系良好,回归方程为y=16.357x+0.007,r=0.9997(n=3),平均加样回收率为97.09%,RSD=2.05%(n=3)。样品中5-FUR前药含量为18.84-18.91μg/ml。结论:该处方设计合理,紫外分光光度法操作简便、稳定性好、结果可靠,可作为IL-2靶向脂质体中5-FUR前药含量测定方法。
Objective:Selection ofthe optimal formulation for IL-2 targeting liposome and determination ofcontent for 5-FUR.Methods:orthogonal test method was used to selection the optimal formulation,UV spectrophotometry was used to determine the 5-FUR prodrug content,and the detection wavelength was at 267 nm.Results:The optimal formulation showed Drug/EPC 1∶10,EPC/Ch 3∶1.The absorbency and concentration of 5-FUR prodrug had good linearity in the range of 2.0~20.0μg/mL.The regression equation was y=16.357x+0.007,r= 0.999 7(n=3),The 5-FUR prodrug content was 18.84~18.91μg/ml.The average recovery rate was 97.09%,and RSD=2.05%(n=3).Conclusion:Formulation was designed reasonablely.The method is simple,stable and reliable,which can be used to determine the 5-FUR prodrug in IL-2 targeting liposome.
