嘌呤受体P2Y_2在涎腺腺样囊性癌的表达及ATP对其细胞生长的抑制

摘要 目的研究嘌呤受体P2Y2在涎腺腺样囊性癌非高转移细胞株(SACC-83)和高转移细胞株(SACC-LM)的表达,及其激动剂ATP对二者细胞生长的抑制作用。方法采用免疫细胞化学法检测P2Y2受体在SACC-83和SACC-LM细胞株中的表达;采用MTT法检测不同浓度ATP(0.1mM/L、0.2mM/L、0.4mM/L、0.8mM/L和1.6mM/L)在不同作用时间点(24h、48h和72h)对SACC-83和SACC-LM细胞生长的抑制作用。结果 SACC-83和SACC-LM细胞中P2Y2均呈阳性表达,在细胞膜的染色较胞浆深,其表达在两细胞株中无明显差异。ATP对SACC-83和SACC-LM细胞的生长均有明显的抑制作用(P<0.05),并且对SACC-LM细胞的抑制作用强于对SACC-83细胞的抑制作用,其抑制作用与药物浓度和作用时间呈正相关。结论不同转移能力的涎腺腺样囊性癌细胞SACC-83和SACC-LM的胞膜和胞浆中均有嘌呤受体P2Y2表达,ATP对二者细胞生长有显著抑制作用。 Objective To investigate the expression of P2Y2 receptor in salivary adenoid cystic carcinoma cell lines (SACC-83 and SACC-LM) and the inhibitive effect of ATP on the cell growth.Methods The expression of P2Y2 receptor on SACC-83 and SACC-LM was measured by immunocytochemical assay. The inhibitive effect of ATP on the cell proliferation was determined by MTT method.Results The expression of P2Y2 was observed both in SACC-83 and SACC-LM cell lines. The cell proliferation of SACC-83 and SACC-LM was inhibited by ATP,and markedly in SACC-LM in dose-time-dependent manner.Conclusion There was the expression of P2Y2 in both SACC-83 and SACC-LM and ATP could inhibit the cell proliferation in both of them.
