双时相rhG-CSF对大鼠外周血CD34^+细胞动员的研究

摘要 目的观察双时相重组人粒细胞集落刺激因子(recombinant human granulocyte colony-stimulating factor,rhG-CSF)对新生大鼠外周血CD34+细胞的动员作用,掌握双时相注射rhG-CSF后外周血CD34+细胞的变化趋势。方法正常7日龄新生大鼠分为3组。A组皮下注射rhG-CSF 50μg/(kg.d),连续注射5 d(第0天至第4天),流式细胞仪测定外周血白细胞中CD34+细胞的比例(CD34+/WBC),当CD34+细胞比例下降3 d后继续给予皮下注射rhG-CSF 50μg/(kg.d),再连续注射5 d;B组皮下注射rhG-CSF 50μg/(kg.d),连续注射5 d(第0天至第4天);C组注射相同剂量的生理盐水,流式细胞仪测定外周血白细胞中CD34+细胞的比例(CD34+/WBC),观察CD34+细胞比例的变化趋势。结果A组外周血CD34+/WBC比例在第4天时达高峰,第5天直线下降,随后1周基本维持在正常值4~7倍的稳定水平。结论双时相rhG-CSF动员能够使外周血CD34+/WBC比例首先达到较高水平,随后1周时间维持在正常值4~7倍的稳定水平。 Objective To explore the effect of double phase rhG-CSF on peripheral blood CD34+ cells in neonatal rats,and investigate the kinetics of peripheral blood CD34 ^+ cells after double phase injection of rhG-CSF.Methods Three groups of postnatal day 7 rat pups were included: Pups in group A were received rhG-CSF(50 μg/(kg.d),subcutaneously) every day for five days(day 0 to day 4),the peripheral blood were collected for CD34^+/WBC count.When CD34^+/WBC descended for three days,five days' rhG-CSF were given again.Pups in group B were injected with rhG-CSF(50 μg/(kg.d),subcutaneous) for five days(day 0 to day 4) once.Pups in group C were received normal saline injections of similar volume for five days.The peripheral blood CD34+/WBC were counted by flow cytometry and kinetics were studied.Results The peripheral blood CD34^+/WBC reached a peak on day 4 in group A,and decreased sharply on day 5,then kept a stable level of 4 to 7 fold the normal level a week thereafter.Conclusions Double phase rhG-CSF can effectively produce the peripheral blood CD34^+/WBC to a peak then keep a stable level of 4 to 7 folds of the normal level for a week.
