摘要
目的:探讨体外模拟腹腔镜二氧化碳(CO2)气腹环境对人卵巢癌细胞株SKOV3生长转移相关因子表达的影响及CO2气腹环境作用后SKOV3对顺铂(DDP)敏感性的变化。方法:将体外培养的处于对数生长期的SKOV3细胞分为4组,即CO2组、DDP组、CO2+DDP组及对照组。用实时荧光定量PCR(Real-timePCR)法及免疫细胞化学法检测各组细胞的PCNA、VEGF和CD44v6mRNA和蛋白的表达。结果:(1)CO2气体与顺铂间无交互作用(P>0.05);(2)CO2处理组肿瘤细胞的PCNA、VEGF和CD44v6mRNA和蛋白表达均明显升高(P<0.01);(3)DDP组与对照组、CO2+DDP组与CO2组比较,DDP处理组肿瘤细胞的PCNA、VEGF和CD44v6mRNA和蛋白表达均明显降低(P<0.01,P<0.01)。结论:CO2环境能促进人卵巢癌细胞株SKOV3生长转移相关因子表达;DDP对肿瘤细胞生长转移因子的表达有抑制作用;CO2环境作用后,肿瘤细胞对DDP敏感性的变化不明显。
Objective:To investigate the effects of laparoscopic carbon dioxide(CO2) pneumoperitoneum environment on the expressions of the growth and metastasis-correlating factors in human ovarian carcinoma cell(SKOV3) and tumor cells sensitivity to cisplatin.Methods:Human ovarian carcinoma cells(SKOV3) in the logarithmic phase were divided into four groups in vitro:CO2 group,DDP group,CO2 plus DDP group and control group.The mRNA and protein expressions of PCNA,VEGF and CD44v6 were analyzed by real-time fluorescence quantitative PCR(Real-time PCR) and immunocytochemical staining.Results:There was no interaction between carbon dioxide and cisplatin(P〉0.05).The mRNA and protein expressions of PCNA,VEGF and CD44v6 were significantly higher in the CO2 treated groups(P〈0.01).The mRNA and protein expressions of PCNA,VEGF and CD44v6 were significantly lower in the DDP treatment groups than those in the non-DDP-treatment groups(P〈0.01,P〈0.01).Conclusion:Carbon dioxide(CO2) promotes the expressions of the growth and metastasis-correlating factors in human ovarian carcinoma cells(SKOV3),while DDP inhibits the expressions.CO2 has no effect on tumor cells sensitivity to DDP.
