摘要
目的探讨血管生长素(ANG)基因与中国汉族人群糖尿病周围神经病(DPN)遗传易感性的关系。方法采用病例-对照研究方法 ,对129例DPN患者和268例健康对照者,应用基因测序技术,检测ANG基因外显子区序列,分析比较DPN患者和健康对照者之间基因型和等位基因分布频率的差异。结果①PCR扩增产物片段长度为736 bp,包含ANG基因整个外显子区和5′侧翼区,DPN组未检测出ANG基因突变;②DPN组和健康对照组的ANG基因外显子区单核苷酸多态性rs11701的基因型(T 442T和T 442G)和等位基因频率(442T和442G)比较后,差异无统计学意义,P>0.05;③按性别分层后,DPN组和健康对照组的ANG基因外显子区单核苷酸多态性(SNP)rs11701的基因型和等位基因频率比较后,差异均无统计学意义,两组P>0.05。结论 ANG基因与中国汉族人群DPN遗传易感性无关。
Objective To investigate the relationship between the angiogenin (ANG) gene and the ge- netic susceptibility of diabetic peripheral neuropathy (DPN) in a Chinese Han population. Methods The sequence of the exons of the ANG gene was detected with a gene sequencing technology using a case-control study in 129 patients with DPN and 268 healthy controls. Results (1)The length of PCR amplified product fragments was 736 bp, including the ANG gene in the entire exon region and 5flanking region, and no ANG gene mutations were detected in the DPN group. (2)After comparing the genotype (T 442T and T 442G) and allele frequency (442T and 442G) of the single nucleotide polymorphism (SNP) rs11701 in the exon region of ANG gene between the DPN group and the control group, there was no statistically significant difference (P 〉0. 05). (3)After comparing the genotype of SNP rsll701 in the exon region of ANG gene with the allele frequency between the DPN group and the healthy control group when they were stratified by sex, there was no statistically significant difference between the 2 groups (P 〉 0. 05 ). Conclusion ANG gene is not asso- ciated with the genetic susceptibility of DPN in Chinese Han population.
