灌注法提取大鼠胰岛及其体内外分泌功能的实验研究 被引量:1

摘要 目的:评估胶原酶灌注法提取大鼠胰岛体内外分泌功能。方法:Wistar大鼠以质量浓度0.05%V型胶原酶胆管灌注提取大鼠胰岛。DTZ、AO/PI染色计数及纯度、活性。葡萄糖刺激试验及体内肾被膜下移植检测其体内外分泌活性。结果:以0.05%的胶原酶提取大鼠胰岛有良好体外分泌活性,胰岛移植可使糖尿病大鼠血糖水平获不同程度下降,部分血糖水平正常。结论:灌注法提取胰岛数量多、活性良好,有良好葡萄糖刺激反应性及移植活性。 Objective: To assess the secretional functions of rat islets in intro and vivo isolated by collagenase perfusion method. Methods : Islets of Wistar rats were isolated by intraductally injecting type- V collagenase of the concentra- tion (w/v) of 0. 05%. The viabilities and functions of islets in vitro were counted by DTZ and AO/PI staining. The in-vivo secretion activity was detected by glucose stimulation test and the in-vivo transplantation under the kidney capsule Results : The secretional activities of the islet in vitro isolated with collagenase of the concentration (w/v) of 0. 05 % was in good condition, the level of blood glouse in some diabetic rats with transplantation of islet was reduced to some degree, but part of them reduced to the nomal level. Conclusions: Islet isolation by perfusion is of great quantities and good activities, which are with good glucose stimulating response and the activity of transplantation.
