摘要
目的:探索乙腈沉淀法去除人血清中高丰度蛋白的效果。方法:应用不同体积的乙腈对血清样品进行处理,Tric ine-SDS-PAGE电泳检测去除高丰度蛋白的效果。结果:乙腈沉淀法能在去除血清高中丰度蛋白的同时收获更多的10 kD以下的小分子量蛋白,2倍体积乙腈结合一定浓度的TFA能有效地去除血清高中丰度蛋白。结论:乙腈沉淀法能去除血清中高丰度蛋白,是一种分离血清部分小分子量蛋白质简便、经济、有效的前期处理方法。
Objective:To explore the effect of acetonitrile precipitation on eliminating high-abundance proteins in human serum.Methods:Elimination of serum high-abundance proteins were practiced with different volume acetonitrile precipitation.Results:The method of acetonitrile precipitation could eliminate the vast majority of high abundance proteins in serum and gain more proteins of low molecular weight.2 times volume of acetonitrile combined with 0.1% TFA could more effective eliminate highly abundant proteins.Conclusion:Acetonitrile precipitation can eliminate high abundant proteins in human serum,which is a convenient,economic,effective method of eliminating micro-molecular protein in human serum.
