用“VRA-GLcNAc”作为底物测尿NAG的活性

摘要 目的:介绍新一代用"VRA-GLcNAc"作底物测尿NAG活性的方法。方法:在pH4.6条件下用二步终点法测定尿中NAG,并做了方法学评估。结果:该试剂的线性可达200 U/L,平均回收率101%。批内和批间变异系数分别为CV2.3%(低值)和1.8%(高值),与8.1%(低值)和6.2%(高值)。与经典的PNP-NAG法比较,r=0.9968,相关良好。结论:液体VRA-NAG测定尿NAG具有灵敏度高,操作简便,结果可靠的特点,它比以前的底物更能满足临床的要求。 Objectire:To describe a new chromagenic substrate and assay for determing NAG activity in human Urine.Methods:Use two step endpoint pH4.6 assay the enzyme activity in urine.Results:The method was in the range of 200 U/L.The average rate of recovery was 101%.The within and between ran CV was 2.3% and 1.8% and VC 8.13% and 6.2%.Comparing this method(Y) with PNP-NAG(X) obtained the regression equation and relative Coeffecinnet Y=1.5019x+0.1268,r=0.9968.Conclusion:This method is simple,sensitive and dependable and suitable for clinical application.
