摘要
目的:观察益气化瘀方对良性前列腺增生模型大鼠血管内皮生长因子(VEGF)、内皮抑素(endostatin)的影响。方法:50只SD大鼠去势7天后,皮下注射丙酸睾酮5mg/kg.d-1,同时给予益气化瘀方、泽桂癃爽混悬液灌胃,30天后股动脉放血处死大鼠,摘取前列腺,测量前列腺体积、计算前列腺指数(PI),免疫组化法检测前列腺组织VEGF、endostatin表达。结果:与正常组比较,模型组大鼠前列腺体积、前列腺指数明显增大(P均<0.01),VEGF表达显著增高(P<0.01),endostatin表达明显降低(P<0.05);与模型组比较,各给药组前列腺体积、前列腺指数减小,VEGF表达降低,endostatin表达增高(P<0.05或P<0.01);与泽桂癃爽组比较,益气化瘀方中、高剂量组前列腺体积、前列腺指数明显减小,VEGF表达明显降低(P<0.05),endostatin表达明显增高(P<0.05)。结论:益气化瘀方抗前列腺增生的作用机制可能与调节VEGF、endostatin表达有关。
Objective:To observe the effects of yiqi huayu prescription on VEGF and endostatin expression in rats with benign prostatic hyperplasia.Methods:50 rats were intracutaneous testosterone propionate after being castrated 7 days to make prostatic hyperplasia models,than they were randomly divided into 5 groups,model,positive and yiqi huayu low,middle,high dose groups,10 rats were taken as normal group,each group was given the corresponding medicine for 30 days.Rats' prostates were extracted after being killed to determine prostate volume,count prostate index,VEGF and endostatin expression in rats' prostatic tissue were detected by immunohistochemical method.Results:To compare with the normal group,the rats' prostate volume,prostate index and VEGF expression were obviously higher in the model group,endostatin expression in rats' prostatic tissue was obviously lower.To compare with the model group,the rats' prostate volume,prostate index and VEGF expression were obviously lowered in each group with medicine and endostatin expression in rats' prostatic tissue was increased.To compare with positive(zhegui longshuang liquid)group,the rats' prostate volume,prostate index and VEGF expression were obviously lower in the middle and high dose groups,endostatin expression was significantly elevated in the two groups.Conclusion:Yiqi huayu prescription can effectively inhibit the prostatic hyperplasia,regulating the expression of VEGF and endostatin expression maybe related to its mechanism.
