电针对脓毒症大鼠肝组织血流量和脂质过氧化的影响 被引量:8

Effects of electroacupuncture on hepatic blood flow and lipid peroxidation in septic rats

摘要 目的:研究电针对脓毒症大鼠肝组织缺血引起的脂质过氧化损伤的保护作用。方法:雄性SD大鼠48只,采用盲肠结扎穿孔术(CLP)制备大鼠脓毒症模型,随机分为假手术组(A组)、模型组(B组)、模型+电针足三里组(C组)、模型+电针非穴组(D组)、迷走神经切断再造模组(E组)和迷走神经切断后造模再电针组(F组),每组8只。E组、F组先切断腹腔迷走神经再施行CLP术。C组、F组持续针刺双侧"足三里"穴1h(2mA,2/100Hz);D组采用相同频率和强度刺激非经非穴("足三里"穴外侧旁开0.5cm)。各组大鼠于CLP术后6h,用激光多普勒血流仪测定肝组织血流量(HBF);检测血浆谷丙转氨酶(ALT)活性;测定肝组织丙二醛(MDA)含量、黄嘌呤氧化酶(XOD)活性以及组织含水率。结果:CLP术后6h,C组肝组织血流量(56.97±11.95)U显著低于A组(80.12±19.57)U,高于B组(42.61±10.97)U、D组(44.53±9.23)U、E组(30.05±4.46)U和F组(30.46±6.38)U(均P<0.05);而MDA含量、XOD和ALT活性及肝组织含水率,C组显著高于A组,低于其他各组(均P<0.05);F组和E组的ALT、MDA、XOD及组织含水率均高于D组(均P<0.05),而肝血流量则低于D组(P<0.05);F组与E组上述指标的差异均无统计学意义(均P>0.05)。结论:电针"足三里"穴能改善脓毒症大鼠肝组织缺血,抑制脂质过氧化,减轻肝组织水肿和功能损害,其机制可能与迷走神经的完整性有关。 Objective To investigate the protection effects of eleetroacupuneture on injury of lipid peroxidation induced by liver isehemia in septic rats. Methods Forty-eight male SD rats were subjected to sepsis induced by cecal ligation and puncture (CLP), and were randomly divided into a Sham operation group (group A), a CLP model group (group B), a CLP model plus electroaeupuncture at "Zusanli" (ST 36) group (group C), a CLP model plus eleetroacupuncture at the shame acupoint (group D), a vagotomy plus CLP model group (group E) and CLP model plus electroacupuncture group after vagotomy (group F), 8 rats in each group. CLP was performed in group E and group F after the abdominal vagotomy. Bilateral "Zusanli"(ST 36) points and the shame aeupoint were electroaeupunctured (2 mA, 2/100 Hz) for 1 hour in group C, group F and group D, respeetively. The hepatic blood flow (HBF) was detected by a laser-doppler flowmetry at 6 h after CLP. The plasma activity of alanine aminotransferase (ALT) was also determined and specimens of liver were harvested for evaluation of malondialdehyde (MDA), xanthine oxidase (XOD) and assessment of the rate of water content. Results The blood flow of the liver was (56.97 ±11.95) U in group C which was significantly lower than (80. 12±19.57) U in group A but higher than (42.61± 10. 97) U in group B, (44. 53±9.23) U in group D, (30. 05±4.46) U in group E and (30.46±6.38) U in group F (all P〈0. 05) 6 h after CLP. Meanwhile, the levels of MDA, XOD, ALT and the rates of water content in liver in group C were all significantly higher than those in group A, but lower than those in the other four groups (all P〈0.05). The levels of MDA, XOD, ALT and the rates of water content in liver in group E and group F were all significantly higher than those in group D (all P〈0. 05), D (P〈0.05), and with no significant differences in all while the blood flow of the liver lower than that in group above measurements between group E and group F (all P〉0. 05). Conclusion Electroacupuncture at "Zusanli" (ST 36) can promote hepatic blood flow, inhibit lipid peroxidation and alleviate hepatic edema and dysfunction in septic rats, which might be related with the completeness of cranial nerve.
