鼠脂肪来源干细胞自体皮下移植的研究

摘要 目的 观察大鼠自体脂肪来源干细胞皮下移植成活的情况.方法 将60只Vistar大鼠随机分为3组,每组20只.实验组,取自体腹股沟区皮下脂肪组织,用Ⅰ型胶原酶消化法提取脂肪来源干细胞并进行传代培养;用脂质体法对细胞进行eGFP(增强型绿色荧光蛋白)转染标记.实验对照组,细胞转染标记后,利用G418使其细胞灭活.空白对照组,注射生理盐水.分别将其注射移植至大鼠自体耳郭皮下层,在注射后第2、4、6周时进行冰冻切片观察,并定量分析所形成组织量的转归情况.结果 实验组切片可见明显的绿色荧光组织层存留,其体积在各个时间点无统计学意义(P〉0.05).结论 将单纯自体脂肪来源千细胞移植于耳郭皮下组织,可以存活,并形成均一稳定的组织结构. Objective To observe the resuh of subcutaneous autotransplantation of mouse's ASCs. Methods Sixty Vistar mouse were divided randomly into 3 groups (20 mouse for each group). In the experimental group, fat tissue was taken from inguinal region and was digested by collagenase Ⅰfor harvesting the ASCs. The ASCs were subcuhured and labeled with eGFP transfection at the second or the third passage. In the control group, the ASCs were killed by G418 after transfection by eGFP. In the blank control group, the ASCs were treated with normal saline. They were injected into the auricle of mouse subcutaneously separately and the resuhs from frozen sections were observed at 2, 4, 6 weeks. Results The new formed tissue labled by eGFP was found in the subcutaneous tissue. Conclusion ASCs can survive after subcutaneous autotransplantation with standard tissue formation.
