摘要 目的:评价小檗碱衍生物HB-13对HaCaT细胞分泌IL-10的影响。方法:采用四甲基偶氮唑盐(MTT)法筛选HB-13的细胞安全浓度;以P物质刺激HaCaT细胞,加入不同浓度的HB-13分别孵育12、24、48 h后,ELISA法及荧光定量PCR法检测培养上清液中IL-10水平及细胞内IL-10mRNA的表达。结果:HB-13浓度为2.5、5、10μg/mL时对HaCaT细胞的毒性作用不显著,但可促进P物质诱导IL-10的分泌,促进作用随时间的延长而增强(P<0.05)。结论:HB-13的抗炎作用部分可能是通过促进细胞因子IL-10的分泌而实现的。 Objective: To evaluate the effect of berberine derivative HB - 13 on the secretion of IL - 10 induced by substance P in cultured HaCaT cells. Method: The eytotoxicity of HB - 13 on HaCaT cells was assessed by MTT method. After HaCaT cells was stimulated by substance P, and cultivated with HB - 13 for 12, 24, 48 h, the level of IL - 10 in supernate and the expression of intra - cellular IL - 10 mRNA were detected by ELISA and real - time quantitative PCR. Results: 2.5,5,10 μg/mL HB- 13 had no cytotoxicity on HaCaT cell line, but promoted the IL - 10 secretion induced by substance P, the amotmt of secretion was time dependent. Conclusion: The anti - inflammatory effect of HB - 13 may be partly accomplished by inducing the production of IL- 10.
