反相高效液相色谱法测定卫矛科植物韧皮部中雷公藤红素含量 被引量:4

摘要 提出了反相高效液相色谱法测定卫矛科植物韧皮部雷公藤红素的方法。样品用索氏提取法,以氯仿为提取剂于75℃提取。提取液以Kromasail C18色谱柱(250 mm×4.6 mm,5μm)为分离柱,以乙腈和0.005 mol.L-1磷酸溶液以体积比80比20混合的溶液为流动相,在425 nm波长处进行分光光度测定。雷公藤红素的质量浓度在0.021 32-0.106 6 g.L-1范围内与其峰面积呈线性关系。用标准加入法测得平均回收率为97.2%。方法用于18种卫矛科植物韧皮部中雷公藤红素含量的测定,腺蕚南蛇藤中雷公藤红素含量最高为1.41%。 RP-HPLC was applied to the determination of tripterine in phloem from celastraceae plants.The sample was treated by Soxhlet extraction with chloroform as solvent at 75 ℃.The extract obtained was separated on Kromasail C18 column(250 mm×4.6 mm,5 μm) with a mixture of acetonitrile and 0.005 mol·L-1 phosphoric acid solution(mixed in the ratio of 80 to 20) as mobile phase and the content of tripterine was determined spectrophotometrically at the wavelength of 425 nm.Linear relationship between the values of peak area and mass concentration of tripterine was obtained in the range of 0.021 32-0.106 6 g·L-1.The value of average recovery found by standard addition method was 97.2%.The proposed method was used in the analysis of 18 kinds of celastraceae plants,and the highest content(1.41%) of tripterine was found in celastraceae orbiculatus.
