木糖醇对糖尿病模型大鼠骨形态计量学的影响

摘要 目的:评价木糖醇(Xy)对糖尿病(DM)模型大鼠骨组织形态计量学的影响。方法:48只♂Wistar大鼠随机分为糖尿病对照(DC)组、DM+10%Xy组、DM+20%Xy组、正常对照(NC)组、NC+10%Xy组、NC+20%Xy组,前3组用四氧嘧啶制作糖尿病大鼠模型且造模成功。各组喂养相应药物12周后处死,留取血及骨标本进行血清总钙(Ca)、磷(P)、碱性磷酸酶(ALP)、骨钙素(BGP)及骨形态计量学指标平均骨小梁板密度(MTPD)、平均骨小梁板间隙(MTPS)、四环素双标记线间的平均距离(DDL)、骨矿化沉积率(MiAR)、骨组织水平的形成率(Svf)测定。结果:与DC组比较,DM+20%Xy、DM+10%Xy组血糖水平显著更低;与NC组比较,DC组血清中ALP和BGP更低;与DC及DM+10%Xy组比较,DM+20%Xy组MTPD水平更高(P<0.05);与NC组比较,DC组MTPS水平更高(P<0.05);与DC组比较,DM+20%Xy和NC组DDL、MiAR、Svf水平更高(P<0.01)。结论:DM大鼠出现骨矿化减少,骨小梁稀疏等骨形态计量学异常;Xy能改善骨组织形态计量学特性,可能对预防和治疗骨质疏松有一定作用。 OBJECTIVE: To evaluate the effects of xylitol (Xy) on bone histomorphology of diabetic mellitus (DM) rats.METHODS: 48 male Wistar rats were randomly divided into diabetic control (DC) group,DM+10%Xy group,DM+20%Xy group,normal control (NC) group,NC+10%Xy group,NC+20%Xy group.Diabetic models of former three groups were induced by alloxan.All groups were given other drugs for 12 weeks and sacrificed.Blood samples and bone samples were collected to determine the level of calcium,phosphorus,ALP,BGP and bone histomorphormetric index,such as mean trabecular plate density (MTPD),mean trabecular plate space (MTPS),distance between double tetracycline labeling (DDL),bone mineral apposition rate (MiAR) and Svf.RESULTS: The level of blood glucose in DM+20%Xy group and DM+10%Xy group were significantly lower than in DC group;The level of ALP and BGP in serum of DC group were lower than in NC group;the level of MTPD in DM+20%Xy group was higher than in DC group and DM+10%Xy group (P0.05);the level of MTPS in DC group was higher than in NC group;the level of DDL,MiAR and Svf in DM+20%Xy group and NC group were higher than DC group (P0.01).CONCLUSION: These results show the abnormality of bone histomorphometric parameters in DM rats.Xylitol can improve bone histomorphometric properties and may prevent osteoporotic changes in diabetic rats.
