苦豆碱对急性期溃疡性结肠炎小鼠的治疗作用 被引量:9

Effects of aloperine on dextran sulfate sodium-induced acute ulcerative colitis in mice

摘要 目的:研究苦豆碱(aloperine)对葡聚糖硫酸钠(dextran sulfate sodium,DSS)致小鼠急性期溃疡性结肠炎(ulcerative colitis,UC)的治疗作用及机制。方法:建立DSS诱导的小鼠急性期UC模型,给予苦豆碱(50,25 mg.kg-1)处理,观察各组小鼠的一般状态、疾病活动度指数、结肠组织病理学变化和组织学评分;检测小鼠血清中IL-1β,IL-4和IL-6的含量以及结肠组织核因子(NF-κBp65)的表达水平。结果:苦豆碱能明显改善结肠炎小鼠的一般状况和结肠组织病理学损伤、降低血清IL-1β和IL-6水平、升高IL-4水平、抑制NF-κBp65的表达。结论:苦豆碱能有效地缓解结肠炎小鼠的炎症反应,其机制可能与抑制NF-κBp65表达、调节促炎因子和抗炎因子之间的平衡有关。 Objective:To investigate the therapeutic effects of aloperine on acute ulcerative colitis(UC) induced by dextran sulfate sodium(DSS) in mice,and to explore the possible mechanism.Methods: Acute ulcerative colitis was induced by DSS,and aloperine was given orally for 8 days at the same time.The general situations and histopathological changes were observed during the experiment.Disease activity index(DAI) and histological score were evaluated.The levels of serum IL-1,IL-4 and IL-6 were determined by ELISA.The expression of nuclear factor(NF-κBp65) in colon was detected by immunohistochemistry and Western blotting.Results: Aloperine greatly ameliorated the symptom of UC and histopathological changes as compared with UC control.The DAI and histological score decreased significantly,the levels of serum IL-1β and IL-6 and NF-κBp65 in colon were obviously reduced,and the expression of serum IL-4 increased after aloperine treatment.Conclusion: Aloperine can effectively attenuate DSS-induced acute ulcerative colitis through decreasing NF-κBp65 expression and regulating balance of inflammatory cytokines.
