还原型谷胱甘肽对毒鼠强中毒大鼠心脑的保护作用 被引量:1

Heart and brain protection of reduced glutathione in acute tetramine poisoning rats

摘要 目的:探讨还原型谷胱甘肽(GSH)对毒鼠强(TET)中毒大鼠心脑的保护作用。方法:健康SD大鼠30只,随机分为假手术组、对照组、GSH预处理组,每组10只。通过TET灌胃染毒大鼠制备动物实验模型,染毒前30 min分别给予对照组、GSH预处理组腹腔注射生理盐水及GSH(1 000 mg/kg),观察各组大鼠的心脑组织病理学改变,免疫组织化学方法评价脑组织γ-氨基丁酸(GABA)、神经元特异性烯醇化酶(NSE)的表达水平。结果:GSH能明显延长TET染毒大鼠的惊厥潜伏期、缓解中毒症状、降低中毒大鼠的病死率,增加脑组织GABA的水平、减少NSE释放,减轻染毒大鼠心脑组织损伤。结论:GSH对TET中毒大鼠有保护作用,为进一步的临床使用提供理论依据。 Objective:To investigate the heart and brain protection of reduced glutathione(GSH) in acute tetramine(TET) poisoning rat. Method:Thirty rats were divided randomly and equally into sham-operated,control and GSH-pretreated group(n =10,each group),the sodium chloride or GSH(1000 mg/kg body wt.) were administered in rats by intraperitoneal injection 30 min before the acute toxicity animal model was performed with TET by intragastric(0.2 mg/kg body wt.).The changes of cell pathology in the heart and brain were investigated with HE method.The expression of gamma-aminobutyric acid(GABA) and neurone specific enolase(NSE) in brain of rats poisoned by TET was analyzed with immunofluorescecnce method.Result:GSH could obviously delay poisoned rats onset of convulsion,improve poisoned rats symptoms after acute tetramine poisoning,reduce poisoned rats mortality,augmentation expression of GABA,decrease release of NSE in brains,and attenuate myocardial/brain damage of poisoned rats.Conclusion:GSH can effectively antagonize acute toxicity of tetramine in rat,and open a new possibility for further clinical application.
