摘要
目的:观察蜘蛛香提取物对惊厥小鼠的抗惊厥作用。方法:采用腹腔注射戊四氮(PTZ)建立小鼠惊厥模型,用蜘蛛香乙醇提取物进行抗惊厥治疗,观察蜘蛛香对惊厥小鼠的行为学及脑组织氨基酸类神经递质γ-氨基丁酸(GABA)和甘氨酸(Gly)含量的影响。结果:行为学观察表明,蜘蛛香提取物对小鼠惊厥潜伏期有一定延长作用、对惊厥率的发生无明显影响,但高剂量组能明显降低惊厥小鼠的死亡率。与模型对照组比较,蜘蛛香提取物还可显著提高脑组织GABA的含量。结论:蜘蛛香提取物对小鼠惊厥潜伏期有一定延长作用,可明显降低惊厥小鼠的死亡率,作用机制可能和升高脑组织抑制性神经递质GABA含量有关。
Objective:To observe the effects of the extract of Valeriana jatamansi Jones on the convulsion model of mice,and investigate the pharmacology mechanism.Methods:The convulsion model of mice was made by intraperitoneal injection of pentetrazole(PTZ).Compared with estazolam as the control medicine,convulsive mice were treated by the extract of Valeriana jatamansi Jones.The ethology and amino acid neurotransmitters in the brain such as GABA and Gly were observed.Results:The observation of ethology indicated that the extract of Valeriana jatamansi Jones could prolong the convulsion latent period in some degree,furthermore obviously decreased the death rate of the convulsive mice in hige dose group(P0.05).Compared with model group,the extract of Valeriana jatamansi Jones may have effect on improving the content of GABA in the brain.Conclusion:The extract of Valeriana jatamansi Jones has the distinct effect of decreasing the death rate of the convulsive mice,and may prolong the convulsion latent period in some degree.The mechanisms are related with improving the content of inhibitory neurotransmitter GABA in the brain.
