健脾祛湿方对重症肌无力模型大鼠骨骼肌线粒体及神经肌肉接头处超微结构的影响 被引量:20

The effect of Jianpiqushi Decoction on ultrastructure of skeletal muscle mitochondria and neuromuscular junction in rats with myasthenia gravis

摘要 目的:观察健脾祛湿方对重症肌无力模型大鼠骨骼肌线粒体及神经肌肉接头处超微结构的影响。方法:采用乙酰胆碱受体α亚基97~116肽段制作实验性重症肌无力模型,透射电镜观察大鼠骨骼肌线粒体及神经肌肉接头处超微结构的变化。结果:正常组大鼠肌纤维结构清晰,神经肌肉接头处突触前膜、后膜结构完整,囊泡与线粒体结构亦完好;模型组可见肌纤维稀疏、紊乱,线粒体数目减少,神经肌肉接头处局部皱折紊乱、溶解、消失,突触前膜囊泡减少、线粒体空泡化等超微结构改变;健脾祛湿方治疗后大鼠肌纤维结构较清晰完整,线粒体数量增多、空泡化减少,神经肌肉接头处较模型组皱折增多而规则,突触后膜线粒体空泡化亦减少。结论:健脾祛湿方能改善重症肌无力模型大鼠骨骼肌线粒体及神经肌肉接头处超微结构改变。 Objective:To observe the effect of Jianpiqushi Decoction on ultrastructure of skeletal muscle mitochondria and neuromuscular junction in rats with myasthenia gravis.Methods:The rats were injected in hypo with synthesized peptide 97-116 of AchR αsubunit to duplicate model of experimental myasthenia gravis(EAMG),ultra-structure of skeletal muscle mitochondria and neuromuscular junction of rats were investigated by transmission electron microscope.Results:Muscle fibrous structure of normal group was clear,with clear structure of presynaptic and postsynaptic membrane of neuromuscular junction and vesicle and mitochondria.In EAMG rats,there were sparse mussy muscle fibre and decreased number of mitochondria,part of the winkle in NMJ were mussy,diffluent,disappeared,and vesicle in presynaptic membrane decreased,vacuolization in organelles like mitochondira existed.After administration of Jianpiqushi Decoction,the muscle fibrous structure of rats was clear,with increased mitochondria and decreased vacuolization in muscle and postsynaptic membrane;and with increased and regulated winkle in neuromuscular junction.Conclusion:Jianpiqushi Decoction could improve ultrastructural changes of skeletal muscle mitochondria and neuromuscular junction in rats with myasthenia gravis.
