选择性环氧化酶-2抑制剂的十年浮沉 被引量：4

摘要 1899年问世的阿司匹林及其后百余年相继上市的40余种其他非甾体抗炎药（nonsteroid anti-inflammatory drugs,NSAIDs）,都为亿万患者提供了重要的抗炎、止痛、解热和抗血栓治疗功效,是至今不可替代的一大类药物.20世纪70年代,研究者们认同了John Van的发现,即阿司匹林类药物的主要机制是对环氧化酶（COX）的抑制,切断了花生四烯酸向炎性介质前列腺素（PG）的转化,故而在临床上起到抑制炎性疾病的作用.但如何解释该类药物在发挥治疗作用的同时出现的如胃肠和其他不良反应？
