摘要
目的 EB病毒相关的淋巴母细胞(lymphoblastoid cell lines,LCLs)有较高的端粒酶活性。本实验旨在分析淋巴母细胞成瘤性与端粒酶活性的关系。方法采用端粒重复扩增法(telomericrepeat amplification protocol,TRAP)分别测定淋巴细胞源性肿瘤细胞以及EB病毒相关淋巴母细胞的端粒酶活性,并对淋巴母细胞的成瘤性进行体内体外试验。结果 5株EB病毒相关的LCLs端粒酶活性与细胞传代次数有关,它们都能在免疫缺陷小鼠体内致瘤。结论容易形成集落的细胞系更容易在小鼠体内致瘤,也具有更高的端粒酶活性。
Objective It is believed that lymphoblastoid cell lines(LCLs) associated with Epstein-Barr virus(EBV) are companied with the high telomerase activity.This article is mainly about the relationship of the tumorigenic cell line and telomerase activity.Methods Clone forming test of 5 LCLs were conducted on agar plate and their telomerase activity were analyzed using an in situ telomeric repeat amplification protocol(TRAP).Results 5 LCLs have the different telomerase activity.All of LCLs can lead tumors in NOD-SCID mice.Conclusion Those cell lines with higher telomerase activity are easier to form clone in mice,and can develope into tumor more easily.
