摘要
目的:通过建立简便的酶速率法,对血浆同型半胱氨酸进行直接检测。方法:使用AU2700全自动生化分析仪,采用两点酶速率法,主波长341nm,副波长406nm,温度37℃,两点线性校准。结果:本法线性范围1.0~51.0μmol/L;日内CV为2.0%~3.1%,日间CV为4.1%~5.8%;低、中、高浓度的回收率分别为102.6%,99.4%,95.9%,平均为99.3%;酶速率法与Abbott Axsym免疫分析仪检测法比较,Y=1.018X-0.865,r^2=0.965。结论:酶速率法简便、快速、敏感、可靠,适用于临床实验室常规和自动化分析,值得推广应用。
Objective: To establish a simple enzyme kinetic method for measuring homocysteine in plasma directly. Methods: Two-point enzyme kinetic method was applied to analyze plasma homocysteine by using AU2700 au -tomatic biochemistry analyzer, The assay parameters are as followings: The dominant wavelength was at 340nm, The subsidiary wavelength was at 405nm, Its temperature at 37 ℃, and two-point linear adjustment. Results: The linearity ra -nge was 1.0 - 51.01amol/L; The CVs of within-day were 2.0% - 3.1%, The CVs of between-day were 4.1% - 5.8%; The recovery rate on low, medial and high consistence were 102.6%, 99.4% and 95.9% respectively, The average recovery rate was 99.3%; The enzyme kinetic method was compared with the Abbott Axsym immunoluminous analyzer, Y= 1.018X-0.865, r^2=0.965. Conclusions: The enzyme kinetic method determination of plasma homocysteine was sim -pie, rapid, sensitive and reliable, It was suitable for the routine automatic analysis and to be applied in clinical practice.
