摘要
目的 研究外源性野生型p53(wt p53)基因对宫颈癌HeLa细胞中环氧合酶-2(COX-2)的影响和对HeLa细胞的生长抑制作用.方法 将p53的HeLa细胞按是否转染p53分为实验组和对照组.实验组利用脂质体介导的转染技术将野生型p53基因转染到HeLa细胞中,wt p53的表达使用逆转录-多聚酶链反应(RT-PCR)鉴定,用蛋白质印迹法(Western blot)与RT-PCR检测转染前后COX-2蛋白和COX-2 mRNA表达的变化.CCK-8染色法绘制细胞生长曲线.结果 野生型p53基因转染后HeLa细胞中的COX-2蛋白和COX-2 mRNA表达减少.通过对细胞生长曲线的分析,转染后较转染前生长明显减慢(P〈0.01).结论 外源性wt p53基因表达可抑制宫颈癌HeLa细胞中COX-2蛋白和COX-2 mRNA的表达;wt p53可能通过抑制COX-2对宫颈癌HeLa细胞产生抑制作用.
Objective To investigate the effects of tumor suppressor gene wild type p53 on expression of cyclooxygenase-2(COX-2)and cell growth of cervical cancer cell line HeLa. Methods Lipofection-mediated gene transfeetion method was used to transfect wild-type p53 (wt p53) gene into HeLa cells. The expression of p53 was confirmed by reverse transeriptase-polymerase chain reaction ( RT-PCR )method. We detected the diversify of expression of p53 mRNA and COX-2 mRNA by RT-PCR, the RNA was harvested after transient transfection and stable transfection. The expression of COX-2 protein was detected by Western blot. The inhibition of HeLa cells by wt p53 was observed with CCK-8 kit. Results After transfection of wt p53, the expression of COX-2 mRNA and COX-2 protein in HeLa-p53 cells were decreased; while the expression of p53 mRNA was increased. Growth curve showed that the cell proliferation of HeLa-p53 was slower than that of HeLa cells (P〈0.01), Conclusion Wide type p53 can inhibit proliferation and COX-2 expression of cervical cancer HeLa cells.
