藏药7号降压作用与毒理学研究 被引量:1

Experimental studies on the antihypertensive effect and the toxicity of Zang Yao-7

摘要 目的:研究藏药7号的降压作用及对动物的毒性作用。方法:采用原发性高血压大鼠(SHR)模型,以降压幅度为指标,观察藏药7号的降压作用;急性毒性试验按Bliss法以不同浓度灌胃昆明种小鼠,计算最大耐受量;长期毒性试验将SD大鼠分为4组,分别以等体积的生理盐水和高、中、低3个剂量的藏药7号连续灌胃12周,观察大鼠的生长发育、血液细胞学、血液生化学、组织病理变化及停药2周后上述指标的变化。结果:降压试验结果表明藏药7号具有明显降低SHR大鼠血压的作用,且量效关系明显;急性毒性试验测定小鼠灌胃藏药7号的最大耐受剂量>15g.kg-1;长期毒性试验中藏药7号各剂量组各项指标未见明显异常改变,与对照组相比差异无显著性。结论:藏药7号降压作用明显,且对小鼠和大鼠无明显的毒性反应。 OBJECTIVE To study the antihypertensive effect and the toxicity of Zang Yao-7, METHODS The spontaneously hypertensive rats were used to investigate the antihypertensive effect of Zang Yao-7. To study the acute toxicity of Zang Yao- 7, oral maximal tolerance dose (MTD) was determined for mice by Bliss method. To observe its long-period toxicity, SD rats were randomly divided into 4 groups, and the 3 groups were administrated with Zang Yao-7 by 0. 3 g.kg-1 .d-1 , 0. 75 g.kg. d-1 and 1.5 g.kg-1 .d -1 respectively, the control group was administrated with 0. 9% NS in identical volume. After continuously feeding rats orally for 12 weeks, the rats' appearance, body weight, the of organ/body coefficient weight, hematological and hemato-biochemical parameters were observed. The routine H&E chromoscopy of heart, liver, spleen, lung, kidney, adrenal gland were carried out. The above physiological signs were inspected again 2 weeks after cease of feeding. RESULTS Zang Yao-7 can significantly decrease the blood pressure in SHR, and the dose-effect relationship was significant. The acute toxicity results showed that Zhan Yao-7 caused no death in mice appeared to the oral maximal tolerance dose of Zangyao-7 in mice was bigger than 15 g.kg-1. The long-period toxicity results showed that the rats' weight increased normally, there was no significant difference between the Zang Yao-7 groups and the control group in the coefficient of organ, hematological and hemato-biochemical parameters, and no abnormal changes were found in the pathological examination. CONCLUSION Zang Yao- 7 has significant antihypertensive effect, and it is safe for clinical application.
