Study on the Relationship between Urinary 1-Hydroxypyrene Level and Nucleoplasmic Bridge and Nuclear Bud among Coke Oven Workers

摘要 目的研究焦炉工尿中1-羟基芘水平与外周血淋巴细胞核质桥和核芽发生率之间的关系。方法分别选取110名焦炉工和50名非接触者,使用调查表收集职业暴露史、年龄、性别、吸烟和饮酒状况等信息。碱水解-高效液相色谱法测定尿中1-羟基芘浓度作为个体多环芳烃暴露内剂量;应用外周血淋巴细胞胞质分裂阻滞微核法制备涂片,计数核质桥和核芽发生率,评价外周血淋巴细胞的染色体损伤水平。结果调整了年龄、性别、吸烟和饮酒状况后,尿中1-羟基芘浓度与外周血淋巴细胞核质桥和核芽率均存在良好的相关关系。进一步按尿中1-羟基芘水平将研究对象分成4组,分别为0.13~1.07组(40人)、1.16~6.77组(41人)、6.87~16.26组(39人)和16.47~72.48μmol/mol肌酐组(40人),应用协方差分析校正了不同内剂量组个体的年龄、性别、吸烟和饮酒状况后,发现尿中1-羟基芘水平与核质桥和核芽率具有较好的剂量-效应关系。结论外周血淋巴细胞核质桥和核芽均能较好反映职业多环芳烃暴露所致的遗传损伤效应。 Objective To investigate the relationship between urinary 1-hydroxypyrene level and nucleoplasmic bridges and nuclear buds in peripheral blood lymphocyte of coke oven workers. Methods 110 coke oven workers and 50 control subjects unexposed to occupational PAHs were recruited. Personal information, including occupational history, age, sex, smoking and alcohol drinking, was collected by questionnaire. Urinary level of 1-hydroxypyrene, taking as a personal internal PAH exposure dosage, was measured by HPLC after alkaline hydrolysis. The chromosomal damage of peripheral blood lymphocyte was evaluated with cytokinesis-block micronucleus method. Results After adjusted for sex, age, smoking and alcohol drinking, a good correlation was found between urinary level of 1-hydroxypyrene and frequency of nucleoplasmic bridges and nuclear buds in study subjects, r=0.79 and 0. 65 respectively, both P〈0.01. All subjects were further divided into four groups according to urinary 1-hydroxypyrene levels namely 0.13-1.07, 1.16-6.77, 6.87-16.26 and 16.47-72. 48 μmol/mol Cr groups. After adjusted for age, sex, smoking and alcohol drinking by covariance analysis, significant dose-effect relationships between nucleoplasmic bridges or nuclear buds and urine 1-hydroxypyrene concentration PAHs exposure levels were found. Conclusions Nucleoplasmic bridges and nuclear buds of peripheral blood lymphocyte well reflect the PAH- induced early genotoxicity.
