摘要
目的比较骨髓动员与动员后单个核细胞移植对糖尿病大鼠后肢缺血的疗效。方法糖尿病大鼠后肢缺血模型建立后,将36只大鼠随机分成3组,其中15只于动员后行单个核细胞移植(A组),15只进行骨髓动员(B组),其余6只为空白对照组(C组),不进行骨髓动员和动员后单个核细胞移植。放射免疫法检测大鼠血浆内皮素(endothelin,ET)水平,免疫组织化学法计数毛细血管数目。结果 A组与B组大鼠缺血后肢毛细血管生成的数量均高于C组(P<0.05),但A组大鼠缺血后肢毛细血管生成的数量高于B组(p<0.05)。2种治疗方法均使血浆内皮素水平显著下降(P<0.05),但下降程度差异无统计学意义(P>0.05)。结论骨髓动员与动员后单个核细胞移植均能促进缺血后肢毛细血管的生成,尤以动员后单个核细胞移植效果显著。2种方法均可改善糖尿病大鼠的内皮功能,两者间差异无统计学意义。
Objective To compare the effects of bone marrow mobilization and implantation of bone marrow mononuclear cells on improving the isehemic disorder of hind limbs in diabetic rats. Methods After hindlimb ischemia diabetic rats model was established, The 36 rats were randomized to three groups,15 rats were treated with b6ne marrow mobilization(group B), 15 rats were treated with implantation of bone marrow mononuelear cells (group A), 6 rats were in control group (group C), without any treating. The level of endothelin (ET) was measured by radioimmunoassay. The numbers of capillary were counted through immunohistochemistry staining. Results The numbers of capillary were higher in both group A and group B than in group C ( P 〈 0.05 ). The numbers of capillary in group A were higher than in group B ( P 〈 0.05). The level of ET was similar between group A and B. Conclusion Therapeutic angiogenesis induced by bone marrow mobilization and implantation of bone marrow mononuclear cells could be an effective treatment for ischemic hind limb disease in diabetic rats. The latter was better than the former. The effects of improving endothelial function of the two methods were similar in diabetic rats.
