利用骨髓间质干细胞修复小鼠颅骨缺损的实验研究

摘要 目的探讨小鼠骨髓间质干细胞(MSCs)修复小鼠颅骨缺损的能力。方法分离培养扩增小鼠MSCs,并在其同窝小鼠颅骨上造约5.0 mm圆形缺损,分别植入煅烧骨+藻酸钙(载体组)和MSCs细胞+煅烧骨+藻酸钠(实验组)及空白组,4个月后,取出标本进行病理分析。结果空白组未见骨组织形成;载体组仅见有少量骨组织形成;实验组有较多的新生骨组织形成,并且新生骨和宿主骨有连接。结论小鼠骨髓间质干细胞能促进骨组织形成,具有修复骨缺损的潜能。 Objective To investigate the potential of bone marrow mesenchymal-derived stem cells(MSCs) in treatment of calvarial defect.Methods Mice MSCs were isolated and cultured in osteoinductive medium.Calvarial bone defects about 5.0 mm×5.0 mm were made on each of the same litter of mice,then filled with none(control group),or calcinated bone and alginate calcium(vehicle group),or MSCs plus calcinated bone and alginate sodium(experimental group).All specimens were removed 4 months post-transplantation for histological evaluation.Results No bone was observed in defects without restoration.A few of new bones were found in defects in vehicle group.Many newly formed bone connected with the end of host bone were also found in defects repaired with MSCs with calcinated bone and alginate sodium.Conclusion MSCs with calcinated bone and alginate sodium have the potential for repairing bone defects.
