小鼠着色真菌病模型皮损内Th细胞因子表达水平观察

Observation on the level of expression of Th cytokines in local lesion in mice chromoblastomycosis model

摘要 目的观察小鼠皮肤着色真菌病模型发病过程中Th相关细胞因子基因表达的动态变化,并进一步探讨其发病机制。方法采用卡氏枝孢霉在ICR小鼠皮下接种建立皮肤着色真菌病模型,在不同时间点RT-PCR法检测皮损组织IL-10、IFN-γ的mRNA表达水平。结果皮损局部IL-10基因表达水平逐渐下降,免疫正常组低于免疫抑制组;相反IFN-γ基因表达水平逐渐上升,免疫正常组高于免疫抑制组。结论 Th1与Th2细胞克隆转向可能在着色真菌病发病中起重要作用。动态跟踪Th1与Th2反应的情况可能为皮肤着色真菌病的发展、转归提供有价值的参考。 Aim To detect the variables of gene expression of helper T cell cytokines in mice model of skin chromomycosis for explore the pathogenesis of the disease. Methods A mice model of chromoblastomycosis was established by inoculating subcutaneously with Cladosporium carrioni, and the level of IL-10 and IFN-γ in the mice lesion tissue was examined by RT-PCR. Results IL-10 mRNA level of each group gradually descended,and that of immunoeompotent group was always lower than that of 2 immunocompressed groups. IFN-γ mRNA level of each group gradually increased,and that of immunocompotent group was always higher than that of 2 immunocompressed groups. Conclusion The clonal diversion of Th1/Th2 ceils might play an important role in the pathogenesis of chromomycosis. Monitoring Th1/Th2 cytokines might be valuable to evaluate the development and prognosis of chromomycosis.
