摘要
目的研究二乙氧基乙醇(2-Ethoxyethanol,2-EE)对大鼠睾丸生精细胞中Bax,细胞色素C(Cytochrome C,CytC)以及Caspase-3表达的影响,探讨其在生精细胞凋亡调控中的作用。方法选择Wistar大鼠,分别给予剂量400、800和1200mg/kg的2-EE,连续灌胃7d,对照组以等量生理盐水灌胃,分别于末次染毒后第1和29天处死动物。采用免疫组化法(SP法),观察不同剂量染毒后及自然恢复4周后生精细胞中Bax,CytC以及Caspase-3的表达情况。结果400、800和1200mg/kg染毒7d后,大鼠睾丸生精细胞中Bax,CytC以及Caspase-3的表达水平不同程度的上升,尤其以800mg/kg组最为明显。自然恢复4周后,生精细胞中Bax,CytC以及Caspase-3的表达基本接近对照组水平。结论在本试验条件下,2-EE急性染毒可能通过影响大鼠睾丸生精细胞中Bax,CytC和Caspase-3的释放,调控生精细胞凋亡,与2-EE所致睾丸毒性的可恢复性高度相关。
Objective To investigate the effects of 2-Ethoxyethanol(2-EE) on the expressions of Bax,Cytochrome C(Cyt C) and Caspase-3 in spermatogenic cells of rat testis,and to explore the mechanism of apoptosis regulation in spermatogenic cells.Methods The animal models exposed to 2-EE were treated with 400,800,1200 mg/kg doses of 2-EE by gavage for 7 days,and the recovery models were fed for another 28 days on the basis of exposed animal models,the control group was treated by 0.9% saline.The Bax,Cyt C and Caspase-3 protein expressions in the spermatogenic cells of rat testis were examined by immunohistochemical technique(SP).Results In comparison with the control group,the expressions of Bax,Cyt C and Caspase-3 proteins in spermatogenic cells increased,principally in the group of 800 mg/kg dose.After 28 days recovery,the expressions decreased and were closed to the control level.Conclusion It was suggested that 2-EE could induce the apoptosis of spermatogenic cells by accelerating the release of Bax,Cyt C and Caspase-3,which was related to the recovery of testis toxicity,and it was significant to improve the risk assessment of 2-EE.
