神经生长因子对胰淀素所致大鼠胰岛细胞功能障碍的保护作用 被引量:2

Protective Effect of Nerve Growth Factor on Amylin-Induced Dysfunction in Rat Islet Cells

摘要 目的:评价神经生长因子(NGF)对高浓度胰淀素所致大鼠胰岛细胞活力和胰岛素分泌功能损伤的修复作用。方法:以不同浓度胰淀素(1、5、10μmol/L)孵育原代培养的大鼠胰岛β细胞,制备功能减退的细胞模型,再以不同浓度的NGF(50、100、200μg/L)孵育高浓度胰淀素(10μmol/L)作用过的胰岛细胞,分别设立空白对照组,比较各组细胞对四甲基偶氮唑盐(MTT)还原能力和胰岛素的分泌能力。结果:高浓度胰淀素(10μmol/L)作用后胰岛细胞对MTT还原能力以及基础糖(5mmol/L)和高糖状态(16.7mmol/L)下胰岛素分泌量的影响与对照组相比均明显下降,差异具有统计学意义(P<0.01),细胞模型制备成功。用不同浓度NGF孵育24h后,随着NGF剂量的增加,此受损胰岛细胞胰岛素分泌量(即24h胰岛素分泌量)、高糖(16.7mmol/L)刺激下胰岛素分泌量均明显增加,受损胰岛细胞对MTT还原能力亦逐步增强。结论:NGF可减轻高浓度胰淀素对胰岛细胞活力的抑制作用,并促进胰岛β细胞分泌功能的恢复。 Objective: To evaluate the effect of nerve growth factors (NGF) on the vitality and insulin secretion in injured islet β cells caused by amylin.Methods: Primary culture of rat islet β-cells was incubated with different concentrations of amylin(1 μmol/L,5 μmol/L and 10 μmol/L)for the dysfunction of cell model.Then the damaged islet β cells caused by highdose amylin(10 μmol/L)were incubated with NGF in different concentrations(50 μg/L,100 μg/L and 200 μg/L),and the MTT reduction capacity and the amount of insulin secretion were observed.Results: The MTT reduction capacity and the amount of insulin secretion in basis of sugar(5 mmol/L)and high-sugar states (16.7 mmol/L) were significantly decreased in the 10 μmol/L amylin group compared with those of control group (P 0.01).The injury cell model was established successfully.When the cells were incubated with NGF (50 μg/L) for 24 h,the insulin secretion was dose-dependent increased.The insulin secretion was significantly increased by high glucose (16.7 mmol/L) stimulation (P 0.01);the MTT reduction capacity of damaged islet cells was also gradually increased.Conclusion: NGF can restore the viability inhibited by high concentration of amylin,and enhance the secretory function of islet β cells.
