不同品系小鼠急性胰腺炎并发肺损伤模型的比较 被引量:3

Comparison of Different Strains of Mice Model with Acute Pancreatitis Associated Lung Injury

摘要 目的:比较小鼠的品系对诱导急性重症胰腺炎并发肺损伤模型的影响。方法:将昆明白、ICR及Balb/c3种品系小鼠各24只于各品系内均分为12、20h对照组及12、20h实验组,每组6只。将3种不同品系小鼠各实验组腹腔内注射雨蛙素诱导并建立急性重症胰腺炎模型,雨蛙素(50μg/kg)稀释于生理盐水中,每小时注射1次,连续腹腔注射12h、20h;各相应对照组腹腔注射等量生理盐水。各组末次注射后1h下腔静脉取血测血淀粉酶,取胰腺、肺脏组织行病理学检查。观察不同品系小鼠注射雨蛙素12h、20h后胰腺、肺组织形态学及血淀粉酶水平差异。结果:昆明白12h实验组及ICR20h实验组仅出现胰腺轻微损伤。Balb/c12h、20h实验组及昆明白20h实验组造模成功,胰腺腺泡广泛变性、坏死及空泡化,炎症细胞大量浸润;肺部间质及部分肺泡腔有炎细胞浸润、肺不张、肺泡间隔增宽。各品系内12h、20h实验组与相应对照组血淀粉酶比较差异有统计学意义(P<0.05)。Balb/c12h、20h实验组各测量值比较差异无统计学意义(P>0.05)。Balb/c12h实验组与昆明白20h实验组病理学检测结果比较差异无统计学意义(P>0.05)。结论:Balb/c小鼠对药物反应灵敏,用药量少,更适用于急性重症胰腺炎模型的研究。 Objective: To investigate the influence of different varieties of mice on the model of acute pancreatitis associated lung injury.Methods:The kunming white (KM),ICR and Bacb/c 3 different strains of mice were divided into 12、 20 h control groups and 12 h、 20 h experimental groups(n = 6).A model of severe acute pancreatitis was induced by intraperitoneal injection of caerulein in Kunming white(KM),ICR and Balb/c strains of mice(n = 12 for each strain).Caerulein (50 μg/kg body weight) in 0.9% NaCl was injected at hourly intervals for 12 h and 20 h.The control animals received the same volume of 0.9% NaCl without caerulein (0.2 mL).The animals were killed after the last intraperitoneal injection.The severity of pancreatitis was evaluated in terms of amylase level and histological changes in KM,ICR and Balb/c at 12 h and 20 h.Results:The model of acute pancreatitis complicated with acute lung injury was successfully established in Balb/c 12 h and 20 h experimental group and KM 20 h experimental group.The pancreatic acinar pathology appeared wide variability,vacuolization acinar necrosis and a large number of infiltrating inflammatory cells.Interstitial and part of the alveolar space were of more neutrophils,atelectasis and the gap widened alveolar.The serum amylase was significantly different in each strain for 12 h,20 h and corresponding control group (P﹤0.05);but no difference between Balb/c 12 h and 20 h experimental groups (P 0.05).There were no difference in pathological result between Balb/c 12 h and KM 20 h experimental groups(P 0.05).Conclusion: Balb/c mice were more suitable for the model of severe acute pancreatitis because of sensitiveness of drug reaction and less drug consumption.
