Modified Catheterization of the Lumbar Spinal Subarachnoid Space in Mice

摘要 目的介绍一种改良小鼠蛛网膜下腔置管技术。方法50只雄性C57BL/6J小鼠,随机分为Wu方法组(n=30)和改良方法组(n=20),分别行蛛网膜下腔置管,观察两组小鼠的置管情况和脊髓解剖情况,置管合格的小鼠术后10d检测运动协调功能和吗啡鞘内注射的镇痛作用。结果两组小鼠在术中死亡率、术后运动功能障碍、脊髓损伤、利多卡因试验阳性率、亚甲蓝定位导管位置上的差异无统计学意义(P〉0.10),改良方法组小鼠的导管泄露率、置管失败率、导管回缩/拔出发生率显著低于Wu方法组俨〈0.05)。两组小鼠置管后第10天的运动协调功能以及吗啡鞘内注射的镇痛作用的差别无统计学意义(P〉0.10)。结论该方法简单易行,成功率高,是适合小鼠蛛网膜下腔置管的可靠方法。 Objective To introduce a modified catheterization of the lumbar spinal subarachnoid space in mice. Methods Fifty male C57BL/6J mice were randomly divided into two groups: Wu-method group (n=30) and modified-method group (n=20). The PE-10 catheter was implanted into lumbar spinal subarachnoid space with two different surgical proceduces. The results of catheterization and spinal nectrotom were compared. 10 days post surgery, the qualified mice were used for tesing locomotor function and analgesic effect of morphine following intratheal injection (i.t.). Result There were no significant differences between two groups in intraoperative death rate, motor dysfunction, spinal cord damage or hemorrhage, lidocaine test and methylen blue lication (P〉0.10), ten days post surgery, there were no significant differences in locomotor function or the analgesic effect of morphine i.t. (P〉0.10). Compared with the Wu-method group, a marked lower rate of catheter leakage, catheterization failure and catheter withdrawal or off was observed in Modified-method group (P〈0.05). Conclusions This modified method is simple and reliable with a high success rate, should be suitable for catheterization of the lumbar spinal subarachnoid space in mice.
