液态氟碳纳米粒增强CT成像在兔VX2肝癌模型中的应用 被引量:9

摘要 目的探讨液态氟碳纳米粒增强CT成像对肝癌的诊断价值及相关机制。方法建立兔VX2肝癌模型,制备液态氟碳纳米粒,于肿瘤种植后18天对荷瘤兔进行CT平扫及在不同时间(即刻、1h、3h、6h、24h、48h)进行液态氟碳纳米粒增强扫描。取兔肝实质及肿瘤组织进行Kupffer细胞免疫组化检测及冰冻切片。结果液态氟碳纳米粒对比剂注射后不同时间点肝实质均明显强化,肝实质与瘤灶的相对影像密度比显著增加。病理检测显示正常肝实质内Kupffer细胞吞噬大量液态氟碳纳米粒,而瘤灶内因缺乏Kupffer细胞无纳米粒的聚集。结论液态氟碳纳米粒可以作为肝网状内皮系统特异性对比剂,实现肝实质的被动靶向强化,对肝癌的诊断具有一定的应用价值。 Objective To investigate the diagnostic value and mechanism of lipid perfluorooctylbromide (PFOB) nanoparticles-enhanced CT imaging on on rabbit VX2 liver carcinoma. Methods Models of rabbit VX2 liver carcinoma were established successfully. Lipid PFOB nanoparticles were prepared by a film-ultrasonic method. CT images were obtained before and at different time (0 h, 1 h, 3 h, 6 h, 24 h and 48 h) after administration of lipid PFOB nanoparticles on 18 days after implantation in rabbits. Immunohistochemistry and frozen sections of Kupffer cells were performed on rabbit's liver and tumor tissues. Results Lipid PFOB nanoparticles obviously increased contrast between tumor and normal liver parenchyma. Pathology examination indicated that there are many nanopaticles taken up by Kupffer cells in normal liver parenchyma, but none in tumor for lack of Kupffer cells. Conclusion As a reticuloendothelial system specific contrast agent, lipid PFOB nanoparticles have potential application value in the detection of liver carcinoma.
