RNA干扰技术在蛋白质组学研究中的应用

摘要 人类基因组测序的完成宣告人类已经进入后基因组时代,此后的一个重要任务就是从蛋白质水平对基因的功能进行验证,高通量的蛋白质组(proteome)研究方法应运而生,为筛选有价值的生物蛋白标记提供了有效的方法;而RNA干扰(RNAinterference,RNAi)技术近年来亦已成为研究基因表达功能的强有力工具之一,它可在转录水平对蛋白质的表达进行调控,这两种新兴研究技术的已经互相成为重要的补充。本文将在简介蛋白质组技术、RNAi技术后,对RNAi在蛋白质组学研究中的现状简要作一综述。 Human genome sequencing has been completed at present,declaring the coming of post-genome era.An important task of post-genome era is to annotate genome function from the protein level.The proteomics study technique emerged as the times require providing an effective method for the screening of biomarkers.RNAinterference technique developed recent year has become one of the most powerful tools in molecular biological study,which could regulate the expression of protein at transcriptional level.These two techniques would be important supplementary to each other.In this review,we will give some introduction to both RNAinterference and proteomics study,and then focus on the combined Application of them.
