芪薏益肝煎对小鼠H22肝癌细胞Cyclin D1及Caspase-3表达的影响 被引量:1

Influence of Qiyiyiganjian on expressions of Cyclin D1 and Caspase-3 in mice bearing hepatocarcinoma H22 cells-derived tumor

摘要 目的观察芪薏益肝煎对H22荷瘤小鼠的抑瘤作用,并探讨其作用机制。方法小鼠右腋下皮下种瘤造成实体瘤模型,将30只小鼠分为3组,对照组、中药组和5-Fu组各10只。用药15 d后取瘤称重计算抑瘤率、胸腺指数、脾指数;采用免疫组化技术检测CyclinD1和Caspase-3的表达。结果芪薏益肝煎对小鼠肝癌细胞的生长抑制率为37.28%,5-Fu组为50.48%。Cyclin D1及Caspase-3在3组中均呈阳性表达,统计结果表明,中药组CyclinD1表达减弱,Caspase-3表达增强,与对照组比较差异有统计学意义。结论芪薏益肝煎对肝癌细胞的增殖有明显的抑制作用,其作用机制与Cyclin D1及Caspase-3表达有关。 A/M To observe the antitumor effect of Qiyiyiganjian on H22 tumor cell in mice and explore its mechanism. METHODS After injecting H22 hepatoma cell into their right axila, 30 mice were randomly and equally divided into 3 groups:control group received treatment of normal saline,treatment group treated by Qiyiyiganjian and 5-Fu group treated by 5-Fu. Fifteen days later, inhibiting rate was caculated. Their thymuses and spleens were isolated and weighed to calculate the thymus and spleen indices. Cyclin D1 and Caspase-3 were examined by using immunohistoehemistry straining method in the tumor tissue. RESULTS The inhibiting rate was 37.28 % in hepatoma cells by using Qiyiyiganjian and was 50.48% by using 5-Fu.The expressions of Cyclin D1 and Caspase-3 in 3 groups were positive. The expression of Cyclin D1 was decreased and the expression of Caspase-3 was increased in treatment group. The diferences between treatment group and control group were significant. CONCLUSION The mechanism of Qiyiyiganjian significantly inhibiting effect on ascites hepatoma cells is relative to expressions of Cyclin D1 and Caspase-3.
