摘要
目的:探讨胰岛素样生长因子-1(IGF-1)对冈田酸(OA)诱导的细胞损伤和tau蛋白过度磷酸化的保护作用。方法:模型组以OA40nmol/L作用于SH-SY5Y细胞24h;IGF-1预处理组分别以100、200和400ng/mlIGF-1预处理2h,再加入OA作用24h。倒置显微镜观察细胞形态学变化;MTT法检测细胞活力;Hoechst染色和分光光度法检测Caspase-3活化程度观察细胞损伤;蛋白免疫印迹法检测tau蛋白磷酸化程度。结果:与模型组比较,IGF-1预处理组细胞形态改善,细胞活力增强,Caspase-3活化程度降低,且磷酸化tau蛋白(Ser396)水平下降。结论:IGF-1可能通过抑制tau蛋白过度磷酸化对OA诱导的细胞损伤具有保护作用。
Objective:To investigate the effects of insulin-like growth factor-l (IGF-1) on cell injuries and tau hyperphosporylation induced by okadaic acid(OA).Methods:The experimental groups were designed as follows:①SH-SY5Y culture (control group);②SH-SY5Y exposed to 40 nmol/L OA for 24 hours(OA group);③SH-SY5Y exposed to OA for 24 hours in the presence of 2 hour pretreatment with 100,200 and 400 ng/ml IGF-1(IGF-1 pretreatment groups).The changes of cell morphology were observed by inverted microscope.The viability of cells was detected by MTT.The injuries of cells were examined by Hoechst 33258 staining and the activity of caspase-3.Western-blot was applied to determine the expression of phosphorylation of tau protein.Results:In IGF-1 pretreatment group,the cell morphology was improved,the viability of cells was increased,and caspase-3 activation and hyperphosphorylation of tau (Ser396)were reduced.Conclusion:IGF-1 can protect the SH-SY5Y cells from cell injuries induced by OA by inhibiting tau hyperphosporylation.
