摘要
目的:为探讨热疗后免疫刺激的机制及血液中白细胞介素2可溶性受体(sIL-2R)的变化。方法:将5×106个LLC(LewisLungCarcinoma)肿瘤细胞接种于C57BL/6小鼠的足垫内,制成肿瘤动物模型,以水浴方式行局部热疗37℃(对照组)、43℃、45℃和47℃,30分钟,观察原发灶肿瘤转移情况,并测定血液循环系统sIL-2R水平。结果:43℃和45℃30分钟热疗后原发灶肿瘤生长速度下降,并且体积逐渐缩小,组织学检查可见肿瘤坏死及炎性细胞浸润,sIL-2R水平下降,肺转移率低于对照组。结论:证明有效的局部热疗后肿瘤坏死分解产物可刺激机体免疫系统,使免疫功能增强,并对杀灭肿瘤原发灶及控制肺转移均起着重要作用。
Objective:To study the mechanism of host immunity enhanced by local hyperthermia and the change of solube Interleukin-2 receptor(sIL-2)after tumor local hyperthermia in mice.Methods:In C57BL/6 mouse was established tumor model by injecting 5×10 6 Leweis Lung carcinoma cells into footpad,and local hyperthermia was performed by water bath at 37℃,43℃,45℃ and 47℃ for 30 min respectively in four groups of the mice.The growth of primary tumor,lung metastasis and the level of sIL-2R were evaluated.Results:The growth and the volume of primary tumor of the foot pad decreased,the necrosis of tumor was found by pathologicl histology examination.The rate of lung metastasis and the level of sIL-2R were lower in the group treated by local hyperthemia of 43℃ and 45℃ for 30 min.compared with the control group.Conclusion:It was possible that tumor decomposition products can enhance the host immunity and plays an important role in eradicating the primary tumor and controlling the distant metastasis.
