消症散结法组方对UUO大鼠肾间质纤维化CTGF表达的影响

Clinical Observation of Xiao-zheng San-jie Method on CTGF Loaded by UUO Rat

目的:动态观察消症散结法组方对单侧输尿管梗阻(UUO)所致肾小管间质纤维化大鼠CTGF表达的影响,探讨其作用机制。方法:111只SD大鼠随机分为正常组、假手术组、模型组、对照组、消症散结法组方(肾炎防衰液)大、中、小剂量组。分别在术后第二周、第四周处死动物,采用免疫组化方法检测肾组织中CTGF蛋白表达。结果:随着梗阻时间的延长,模型组大鼠肾组织CTGF表达增加;与模型组各对应时间点比较,消症散结法组方(肾炎防衰液)治疗后可使肾组织中的CTGF表达明显降低(P<0.05)。结论:消症散结法可通过抑制CTGF表达,减轻肾间质纤维化。

Objective： To observe dynamically the effect of Xiao-zheng San-fie method on connective tissue growth factor （CTGF） in the rat with renal interstitial fibrosis caused by unilateral ureteral obstruction （UUO）, and investigate its mechanism of action. Method ： 111 SD rats were randomly divided into following groups, which were normal group, sham operation group, model group, control group and high, medium and low dose group of Xiao-zheng San-fie method. Respectively in the second and fourth week after operation the animal were put to death, and the expression of CTGF in nephridial tissue was detected with immunohistochemical method. Result： With the time of obstruction prolonging, the expression of CTGF increased in model group. Compared with the cor- responding time points in UUO group , Xiao-zheng San-fie method decreased the expression of CTGF in nephridial tissue markedly （ P 〈 0.05 ）. Conclusion ： Xiao-zheng San-fie method significantly alleviates renal interstitial fibro- sis by suppressing the expression of CTGF.
