摘要
目的:探讨复元醒脑汤对高血压性脑出血大鼠脑组织形态学改变的干预作用。方法:将SHR大鼠随机分为假手术组、模型组、安宫牛黄胶囊组、复元醒脑汤组,每组20只,并与WKY大鼠假手术组20只对照。模型组、安宫牛黄胶囊组和复元醒脑汤组在收缩压达到21.3kPa基础上以胶原酶加肝素尾状核注射法诱发脑出血,建立高血压性脑出血模型,其中安宫牛黄胶囊组和复元醒脑汤组于造模后分别给予相应药物干预,连续灌胃3天。两假手术组脑内注射及灌胃均为生理盐水,模型组灌胃为生理盐水。观察各组动物神经缺损行为体征、脑组织含水量及病理形态学改变。结果:SHR模型组与WKY假手术组、SHR假手术组比较,神经缺损行为体征评分升高(P均<0.01),脑组织含水量增加(P均<0.01),出现病理形态学改变;SHR复元醒脑汤和SHR安宫牛黄胶囊两治疗组与SHR模型组比较,神经缺损行为体征评分降低(P均<0.01),脑组织含水量减少(P均<0.01),病理形态学改变明显改善;复元醒脑汤组与安宫牛黄胶囊组比较差异无统计学意义(P均>0.05)。结论:复元醒脑汤可改善高血压性脑出血大鼠脑组织病理形态学改变,减轻脑水肿,改善神经功能缺损。
Objective: To study the effect of Fuyuan Xingnao Decoction on morphology changes of brain tissues in hypertensive cerebral hemorrhage rats. Methods:Spontaneously hypertensive rats (SHR) were randomly divided into sham operation group( SHRS, n = 20 ) , model group ( SHRM, n = 20 ) , Angong Niuhuang capsule group ( SHRTa, n = 20 ) , and Fuyuan Xingnao Decoction group( SHRTf, n = 20) , compared with Wistar Kyoto rat sham operation group( WKYS, n = 20). After the systolic pressure stabilizing in 21.3 kPa, SHRM,SHRTa and SHRTf were induced cerebral hemorrhage by caudate nucleus injection with collagenase and heparin. Then SHRTa and SHRTf were given corresponding drugs via intragastric administration for 3 days after getting into cerebral hemorrhage, while SHRS and WKYS were all given intracerebral injection and intragastric administration with saline, and SHRM was given intragastric administration with saline. Rats of all groups were respectively observed their behavlor and physical signs of neurological defect, brain tissue moisture capacity and morphology changes. Results:Compared with WKYS and SHRS groups ,the grades of SHRM group' s behavior and physical signs of neurological defect and brain tissue moisture capacity increased( P 〈 0. O1 ) , presented pathological changes in brain tissue;SHRTa and SHRTf groups' behavior and physical signs of neurological defect grades and brain tissue moisture capacity lowered as compared to SHRM group (P 〈 0.01 ) , and their pathological changes in brain tissue improved;there is no statistical significant difference in the index changes between SHRTa and SHRTf groups ( P 〉 0.05 ). Conclusion: Fuyuan Xingnao Decoction may achieve the goal of treating hypertensive cerebral hemorrhage through improving pathological changes in brain tissue, relieving cerebral edema, and repairing neurological impairment.
