摘要
我们合成了抑制素α亚单位N末端1-32氨基酸(32肽.P23).[Tyr]-1-32氨基酸(33肽,P33)和37-65氨基酸(29肽,P29)片段。本实验观察这三个片段对排卵和着床影响。在PMSG-HCG诱发小鼠超排卵实验中.P29具有显著的刺激超排卵作用.P29组排卵率是单纯hCG组的两倍(P<0.001)。在着床实验中.利用大鼠双子宫角的特点进行自身对照实验。在胚泡着床前期,对妊娠大鼠一侧子宫角注入100μl生理盐水作为对照组,另一侧注入100μl(300ng)抑制素α亚基片段。结果除P29一例外.均完全阻断胚泡着床(P<0.001)。
Human inhibin α - subunit-N-terminal 1-32 amino acid (peptide 32. P32 ). [Tyr]-1-32 (P33) and 37-65 (P29) were synthesized. In this edperiment. the effects of 3 fragments on ovulation and implantation of embryo were observed. PMSG-hCC-induced superovulation mouse model was used for the testing of ovulation. Among 3 fragmentls. only P20 showed significant stimstimulating effect on ovulation. the ovulation rate in P29 group was twice as much as in control (hCGinduced superovulation rate ) (P< 0. 001 ). For implantation experim,ent.100μl vehicle was injected one side uterus of pregnant rat before implation and the other side uterus received 100 μl (300 ng ) of inhibin frament P29. P32 and P33 respectively. The result showed that except for one case in group of P29. they all absolutely block the implantation of embryo (P<0.01).
