摘要
目的比较研究10种人造矿物纤维(MMMF)对人肺泡上皮细胞(A549细胞)DNA的损伤及修复作用。方法选用日本纤维状物质研究协会提供的10种标准人造矿物纤维(JFM),用单细胞凝胶电泳法,以A549细胞作为靶细胞,测定了细胞DNA链断裂、损伤后修复及DNADNA链间交联。结果10种JFM纤维都可引起不同程度的DNA链断裂、DNADNA链间交联及抑制DNA损伤后的修复功能。综合分析发现:微细玻璃纤维的遗传毒性最强,硅灰石的遗传毒性最低。它们的作用程度均低于温石棉。结论JFM纤维均具有一定程度的体外遗传毒性,但均低于UICC温石棉B。
Objective To study and compare DNA injuries and repair induced by 10 kinds of man
made mineral fibers(MMMF) in human alveolar epithelial cell line(A549 cell).Methods Single
cell gel electrophoresis(SCGE) assay was used to detect DNA strand breaks,DNA DNA
interstrand crosslink and the ability of DNA repair when A549 cells were incubated with 10 kinds
of JFM fibers and UICC chrysotile B respectively.Results All of 10 kinds of JFM fibers could
induce DNA strand breaks,DNA DNA interstrand crosslink and inhibit the ability of DNA
repair,but all of these effects were lower than that of chrysotile and were different among
them.Conclusion All of 10 kinds of JFM fibers had genotoxicity with different degrees in vitro ,
but lower than that of chrysotile B.
