摘要
目的探讨建立硫高铁血红蛋白的测定方法以评价不同高铁血红蛋白形成剂对急性硫化氢中毒的解毒作用。方法采用标准方法、定值参考物进行质量控制。用硫化氢气体饱和高铁血红蛋白生成硫高铁血红蛋白样品,扫描吸收曲线。氰化高铁血红蛋白与硫高铁血红蛋白在各自特征吸收峰540nm、654nm处的毫摩尔消光系数,求得硫高铁血红蛋白百分比浓度。结果质控变异百分率V≤0.4%,各消光系数变异系数CV≤3.0%,平均回收率97.5%±4.1%;重复实验CV≤1.5%,n=10;线性范围0.0%~80.0%,直线回归方程为Y=0.973X+0.158、r=0.999。结论该方法灵敏、准确且简便、快速,适用于临床和科学研究。
Objective Methods The levels of sulfmethemoglobin in blood were measured accurately utilizing standard methods and instrument.Sulfmethemoglobin specimens were prepared from methemlglobin solution saturated with pure hydrogen sulfide.After scanning the optical absorption spectra of HICN and SMHb,the extinction coefficients were obtained under their respective characteristic absorbance peak(540 nm and 654 nm).The concentration of sulfmethenoglobin was then calculated according to the equations deduced from the Bears law. Results Quality control variation rate was: V ≤ 0.4% ;all extinction coefficient: CV ≤ 3.0% ;average recovery rate: 97.5%± 4.1% ;repetition experiments: CV ≤ 1.5% ( n =10);linear range 0%~ 80% ;regerssion equation Y = 0.973 X +0.158, r =0.999. Conclusion The method appears to be a sensitive,accurate and simple technique suitable for clinical and scientific research.
